Multipotent stromal stem cells from normally discarded human placental tissue demonstrate high therapeutic potential

ScienceDaily (May 18, 2012) ? Scientists at Children's Hospital Oakland Research Institute (CHORI) led by Vladimir Serikov, MD, PhD, and Frans Kuypers, PhD, report in the current Epub issue of Stem Cells Translational Medicine that placental stem cells with important therapeutic properties can be harvested in large quantities from the fetal side of human term placentas (called the chorion).

The chorion is a part of the afterbirth and is normally discarded after delivery, but it contains stem cells of fetal origin that appear to be pluripotent -- i.e., they can differentiate into different types of human cells, such as lung, liver, or brain cells. Since these functional placental stem cells can be isolated from either fresh or frozen term human placentas, this implies that if each individual's placenta is stored at birth instead of thrown away, these cells can be harvested in the future if therapeutic need arises. This potential represents a major breakthrough in the stem cell field.

In previous work, Drs. Serikov and Kuypers reported a novel technology to harvest blood-forming stem cells from the placenta to augment cord blood cells. These cells are "siblings" of the cord blood derived stem cells. Cord blood stem cells, unlike embryonic stem cells, have been used for many hundreds of successful bone marrow transplants. These transplants are mainly performed in children, as the amount of cells that can be harvested from cord blood is usually not sufficient for a successful transplant in adults. Adding placental-derived stem cells to the cord blood stem cells could make successful adult bone marrow transplants routinely possible.

The current report demonstrates that placental stem cells have much broader therapeutic potential than bone-marrow transplants, because they are pluripotent -- i.e. able to differentiate into many different cell types -- and they also generate growth factors that help in tissue repair. These cells are shown to integrate into different tissues when transplanted into mice, but like cord blood stem cells, and in contrast to embryonic pluripotent stem cells, they do not form tumor-like structures in mice.

Placental-derived stem cells are often viewed as "adult" stem cells in contrast to "embryonic" stem cells, which are the dominant focus in the stem cell research field. However, this report shows that these fetal stem cells can be harvested in large numbers, and without the ethical concerns attached to the use of embryonic stem cells. These stem cells may thus be a more practical source for regenerative medicine, particularly since, if placentas are routinely saved instead of thrown away, each individual will be able to draw on their own fetal stem cells if future therapeutic needs arise.

Placental stem cells are only 9 months old, and in contrast to adult stem cells, do not need to be reprogrammed to become pluripotent. Placental-derived stem cells have characteristics of young and vigorous cells, including young mitochondria. Future research will be aimed to bring this to the clinic and to test their efficacy in translational therapeutic applications.

Daniel Radcliffe And Jon Hamm Join British Miniseries

Actors will play younger and older version of Russian doctor in four-part dramedy 'A Young Doctor's Notebook.'

What do Don Draper and Harry Potter have in common? They're both about to become doctors.

The Internet was buzzing earlier this month when rumors popped up that actors Jon Hamm and Daniel Radcliffe were set to play the older and younger version of a Russian doctor in a four-part British miniseries set during the end of World War I. On Friday (May 18), Sky Arts, the network set to air the series in 2013, confirmed that we'll indeed get to see Dr. HammRad in action. "A Young Doctor's Notebook" is a dramedy based on a short-story collection by Mikhail Bulgakov, a Russian writer and playwright.

"I have been an obsessive Bulgakov reader for a couple of years now," Radcliffe said in a statement about the television program. "So when the opportunity to become involved in this project came up, I could barely contain my excitement. The book is funny, grotesque and heartfelt in equal measure and I look forward to working with a great group of people to help bring it to life."

Hamm agreed with his new co-star's enthusiasm, saying, "I am thrilled to get the opportunity to work on such rich source material with such fantastically talented people whose work I greatly respect."

The actors will play the same character in different time periods, occasionally interacting with each other. The original rumors were reported by The Sun, which said the show "will incorporate dark humour as the doctor attempts to overcome self-doubts about his competence."

When MTV News spoke to Radcliffe last summer, he explained he's attracted to darker tales because he finds them much more interesting.

"I don't want to make a boring love story about two people who meet and then are happy. That's boring, and that doesn't exist," Radcliffe said. "I also do like playing slightly disenfranchised characters like Arthur is in 'The Woman in Black.' He's someone who's grieving and whose grief has put him to the edge of madness and also has sort of made him ostracize himself from society. I guess I like the dark stuff. I find it more interesting."

Hamm said producer Clelia Mountford and production company Big Talk have been "unabashedly" advocating for the series. "I share their great enthusiasm for bringing something original, dark, funny and moving to light," the actor said. "Also, I have been watching 'Bridget Jones's Diary' on infinite loop, and I think I've finally got this accent thing sorted..."

Slo-mo Microbes Extend the Frontiers of Life

I'm not really sure what finding life on Mars will do for us though. It will show us that life on another planet is possible (something I personally believe already), but other than that knowledge, what will we gain from it? It won't help solve our current environmental, economic of political problems, will it?

Well, the human race can multitask, after all. If that really is a sign of complex life on Mars, and it looks uncannily biological, it will give us our first glimpse of a completely new permutation of life. If its similar to our own, we have immediately given huge weight to innumerable theories, and undermined many more. If its something completely alien, the same applies in reverse. What practical uses we might put the knowledge to are a complete unknown at this point. Maybe it will help cure cancer, maybe
